A single bead lock will only support one side of the tire as a double supports both. A single bead lock can also allow the inside tire bead to slip causing your tire to twist. The tire contact patch is crucial and thus a double is always the best.

A beadlock will make the tire sidewall stiffer.
The more mph you throw at tire, the more the tire is going to grow and change shape. A non-beadlock wheel allows the tire to grow more; but as the tire distorts more to allow this growth, you have less tread on track and the chances increase for the tire to separate from the wheel due to centrifugal force. Any speed in excess of 175 mph greatly increases this risk.
The beadlock not only holds tire to keep it from spinning on the wheel, but also limits the tire growth to keep more tread on the track for increased stability and reduced tire shake. The rim screws do limit tire slippage, but do not affect the tire sidewall and growth as the beadlock would.
Another point to consider about a single vs. double beadlock is the weight. A beadlock will add up to 6 lbs to per double beadlock wheel. If you are using only a single beadlock on a wheel, you will need to compensate the additional 3lbs of beadlock weight on the outside of the wheel with the rim screws and/or wheel weights on the inside of the wheel to keep the rotating weight balanced.
